PROPRIETATILE FARMACOLOGICE
Acidul nicotinic sau niacina este o vitamina hidrosolubila din complexul B (vitamina PP). Intra in componenta coenzimelor NAD (nicotinamid – adenin – dinucleotid) si NADP (nicotinamid – adenin – dinucleotid fosfat), participa la metabolismul lipidelor, aminoacizilor, purinelor, procesul de glicogenoliza si respiratia tisulara. Acidul nicotinic (dar nu si nicotinamida) scade nivelul LDL (Low – density Lipoproteins), colesterolului si trigliceridelor in sange care reprezinta un factor de risc al cardiopatiei ischemice, apolipoproteinei B – principalul component al LDL (Low – density Lipoproteins) si VLDL (Very Low – density Lipoproteins) care sunt factori de transport al colesterolului in sange si lipoproteinei A, o forma a LDL asociata cu riscul cardiopatiei ischemice. Diminueaza esterificarea hepatica a trigliceridelor. Reduce concentratia fibrinogenului plasmatic si creste nivelul catalizatorului tisular al plasminogenului, care reprezinta factorul de inhibitie al lipazelor tesutului adipos. Dilata vasele periferice.

INDICATII TERAPEUTICE
Profilaxia si tratamentul pelagrei; bolile hepatice (hepatitele acute si cronice, cirozele); gastro – intestinale (gastrite, enterocolite, colite), spasmele vasculare, ateroscleroza). Ictusul ischemic (adjuvant). Ulcerele si plagile trenante.

DOZE SI MOD DE ADMINISTRARE
In tratamentul pelagrei la adulti, parenteral cate 10 mg 1 – 2 ori pe zi timp de 10 – 15 zile. In ictusul ischemic 10 mg/zi, intravenos. In tratamentul hipercolesterolemiei si hipertrigliceridemiei cate 1 – 2 g de 2 – 3 ori pe zi. La debutul tratamentului doza va fi de 0,5 g, seara dupa cina. Dozele si frecventa administrarii vor fi corelate in dependenta de efectul obtinut asupra nivelului plasmatic al LDL – colesterolului sau trigliceridelor.

CONTRAINDICATII
Hemoragiile arteriale, glaucomul, diabetul zaharat (dozele mari), guta, hipotensiunea arteriala, ulcerul peptic activ, hipersensibilitatea la Acid nicotinic.

Prezentare, Ambalaj: Solutie injectabila 1% - 1ml N 10.

Pastrare: Lista B. La loc ferit de lumina, la temperatura 15 - 25°C.
