Actiunea farmacologica: N-colinolitic central cu actiune inhibanta preponderent in partea caudala a formatiei reticulate. Are actiune membranostabilizatoare si anestezica locala, inhiba conductilitatea impulsurilor nervoase ale fibrelor aferente si neuronilor motorii, provoaca blocajul reflexelor spinale mono- si polisinaptice. Este probabil ca inhiba eliminarea neurotransmitatorilor prin blocarea canalelor ionilor de calciu si influxului acestor ioni in terminatiunile sinaptice. Scade tonusul muscular crescut, rigiditatea musculara, amelioreaza miscarile musculare voluntare. Are actiune spasmolitica si vasodilatatoare moderata, creste circulatia periferica printr-un mecanism spasmolitic si antiadrenergiC
INDICATII
Bolile neurologice organice asociate cu hipertonus muscular (leziunile sistemului piramidal, scleroza diseminata, mielopatia, encefalomielita s.a.), hipertonusul muscular, spasmele musculare, contracturile musculare, rigiditatea musculara, automatismul spinal, bolile vasculare obliterante (ateroscleroza obliteranta, angiopatia diabetica, trombangiita obliteranta, boala Raynaud, sclerodermia sistemica), sechelele afectiunii inervatiei vasculare (acrocianoza, claudicatia intermitenta angioneurotica), tulburarile posttrombotice ale circulatiei limfatice si venoase, ulcerele trofice gambiene, paralizia spastica (boala Little), encefalopatia asociata cu distonie musculara.
DOZE SI MOD DE ADMINISTRARE
Adulti cate 150-450 mg de 3 ori pe zi p.o. sau cate 100 mg de 2 ori pe zi i.m. sau 100 mg i.v. lent o data pe zi. Poate fi administrat intraarterial (continutul fiolei se dilueaza in 10 ml ser fiziologic). Copii 3 luni-6 ani 5 mg/kg/zi divizata in 3 prize, 7-14 ani 2-4 mg/kg/zi.
CONTRAINDICATII
Miastenia, varsta sub 3 luni, hipersensibilitatea la preparat.
ATENTIONARI SI PRECAUTII SPECIALE DE UTILIZARE
A se testa sensibilitatea la lidocaina. Copiilor li se indica numai formele orale (50 mg). Sarcina (numai in cazuri exceptionale).
